The plates looked like this today:
I chose colonies from all four colors and incubated in liquid LB following the protocol at the beginning of the document, with a modification:
I've added an "LB+DH5α" control that I expect to turn cloudy when incubated to check that the LB was made correctly and not contaminated with any antibiotics or bactericidal impurities, in addition to the "LB only, no bacteria" control that I expect to incubate clear to verify sterile technique and equipment. I've replaced the “LB+chloro” control with a "LB+chloro+DH5α" control that I expect to incubate clear to verify antibiotic potency against ostensibly non-resistant bacteria.
